Glycerophosphorylcholine, abbreviated as GPC, is one of the phospholipid families. It plays an important role in the health of cell membranes, glycerol phosphorylcholine is very rich in breast milk, repairs damaged brain function, improves brain function in healthy people, reactivates the aging brain, as a health food, can promote the biosynthesis of acetylcholine and glycerol phosphorylcholine in the brain, improve people's memory and cognitive ability, and can also effectively treat Alzheimer's disease and other Alzheimer's diseases, known as "brain anti-aging nutrients" in the pharmaceutical community, let's learn about the synthesis and purification method of glycerol phosphorylcholine!
A method for refining glycerophosphorylcholine, comprising the following process;
a. Dissolve the crude glycerol phosphorylcholine in absolute ethanol to obtain the dissolved solution of the reaction system; The mass ratio of the absolute ethanol relative to glycerophosphorylcholine is 61.
b. Add calcium chloride to the dissolved solution of the reaction system in process A, stir to form a calcium salt complex precipitate of glycerol-glycerol-phosphorus-philcholine; The molar ratio of calcium chloride and glycerophosphatidylcholine is 11. The temperature condition of the precipitation reaction is room temperature, and the reaction time is 12 hours.
c. Reduce the reaction solution that stops the precipitation reaction in process B to room temperature, filter it, and obtain a cake; Cakes are washed with absolute ethanol.
d. Put the filter cake of process C into a vacuum drying oven to dry, and obtain the calcium salt of glycerol phosphorylcholine. The purity determination of HPLC method was above 98.5%.
e. Calcium glycerol phosphorylcholine salt is soluble in water, and the water consumption is 10 times the dry weight of calcium glycerol phosphorylcholine salt, and it is stirred and dissolved at room temperature. If it is difficult to completely dissolve after stirring for a long time, you can drop a few drops of hydrochloric acid into the aqueous solution to accelerate the dissolution (controlling the solution to reach the appropriate acidity will help dissolve.) The pH value is between 3~7, and the pH value is preferably 4~6).
f. The aqueous solution obtained in process e is slowly removed by the D001 cation exchange resin column in turn, and the anions are removed by the D201 anion exchange resin column (or D301 anion exchange resin column) to obtain an aqueous glycerol ester solution without calcium ions.
Distillation removes excess water to obtain glycerol phosphorylcholine, which does not contain calcium ions.
The physicochemical structure and interrelated physicochemical properties of phospholipids ensure that they are very beneficial to brain tissue, and often become phospholipids that protect the metabolism of brain tissue. Acetylcholine can help with activities such as memory and learning in the brain. In vivo, choline, a raw material required for acetylcholine biosynthesis, provides the physiological and biochemical properties of glycerol phosphorylcholine through the blood-brain barrier. The Cholinergic Injury Hypothesis argues that the loss of cholinergic properties is related to the severity of Alzheimer's disease.
